Trypsin Enzyme Application

  • For biological medicine production, such as Insulin, GLP-1 drugs.

  • Protein structure and function studies, such as protein mass spectrometry, sequencing, peptide mapping analysis.

Trypsin Enzyme Recommended Method

  • Protein preparation: The protein was dissolved in a solution of 25 mM Tris-HCl, pH 8.0. 

  • Enzyme preparation: The desired enzyme powder was weighed and dissolved in 10 mM HCl to give an enzyme solution concentration of 1-10 mg/ml. 

  • Reaction conditions The amount of enzyme: the target protein (mass ratio) = 1:50~1:1000, the optimum pH is 7.0~9.0.1. 

  • Solution preparation:

  1. Recombinant Trypsin is thawing at room temperature. According to the protein content in COA, take an appropriate amount of trypsin and add HBSS balance solution (or other buffers suitable for cell digestion, if you need to add EDTA, the final concentration is recommended to be 0-1mM, preferably no more than 2mM) The concentration of trypsin used is about 0.1- 0.3mg/ml (the concentration is adjusted according to different cells), and mix gently; this step can be performed at room temperature; 

2. Use 0.22 μm filters to filter the above trypsin solution and transfer it to a sterile container; this step can be performed at room temperature;

3. The filtered trypsin solution should be used directly on the day as required (for example, add 1ml to a T25 flask, digest cells at 37°C)

Trypsin Enzym Transport and Storage Stability

  • Transportation stability: It is transported in an ice pack to keep it active.

  • Storage stability: The lyophilized powder can be stored for at least 24 months at -15°C below.

  • 1 mM hydrochloric acid or 50 mM acetic acid was dissolved and stored at -15°C below, repeated freeze- thaw 10 times, no loss of activity.

  • It is recommended that the storage should not exceed 7 days after dissolution at 2~8℃, and it should not exceed 30 days after storage at -15°C below.
